Późnobarokowy, budowany w latach 1729 - 1744, z piękną, rokokową fasadą. Wnętrze kościoła jednonawowe w układzie centralnym. Kościół wchodzi w sklad barokowego Zespołu Klasztornego Benedyktynek. Zakonnice sprowadzone zostały tu z Torunia 1 listopada 1623 r., prowadziły nauczanie dziewcząt ze środowiska szlacheckiego. Mimo ograniczeń benedyktynki drohiczyńskie prowadziły swą oświatową działalność. W wyniku represji carskich kościół został zamknięty, zakonnice przewiezione do Wilna roku 1856. Wróciły w czasie powstania styczniowego, prowadziły tajną polską szkołę w Drohiczynie, zorganizowały szpital dla powstańców. Aresztowane po upadku powstania przebywały w więzieniu w Bielsku Podlaskim, następnie cztery z nich zesłane zostały na Sybir w roku 1864. Klasztor zburzyli kozacy.W końcu XIX w. klasztor wraz z kościołem były poważnie zdewastowane. Odnowiony w okresie międzywojennym. W latach 1939 - 1941 sowieci przerobili kościół na toalety dla żołnierzy.
