23 października 1623 roku Wojciech Niemira, kasztelan i wojewoda podlaski, ofiarował plac pod budowę kościoła i klasztoru w Drohiczynie oraz dwór i 3 włóki ziemi w Sytkach. Celem fundacji było stworzenie możliwości edukacji dziewcząt z możniejszych rodów szlacheckich na Podlasiu. Donator własnym kosztem wzniósł drewniany klasztor i kościół pw. Wszystkich Świętych. Ważną rolę w sprowadzeniu sióstr odegrał o. Anioł Bidsiński, gwardian drohiczyński ojców franciszkanów. Pierwotna donacja została powiększona dobrami ziemskimi, które odziedziczyła Zofia Kiszko, córka Mikołaja Kiszki, starosty drohickiego. W dniu 1 listopada 1623 r. przybyła ona do Drohiczyna z klasztoru toruńskiego wraz z jedenastoma zakonnicami. Na pamiątkę dnia jej przybycia kościół otrzymał tytuł Wszystkich Świętych. Zofia Kiszczanka w 1630 r. została pierwszą ksienią nowego klasztoru, którego kolatorami byli królowie. Benefaktorami kościoła i klasztoru sióstr, obok władców Polski, byli duchowni, okoliczni ziemianie, szlachta i mieszczanie.
