Kościół późnogotycki z 1520 r., ufundowany i uposażony przez wojewodę litewskiego, Mikołaja Radziwiłła, wybudowany przez Jana Zamoyskiego, przebudowany do stanu dzisiejszego w latach 1899 - 1902. Kościół murowany, jednonawowy, z dwiema kaplicami, wydzielonym prezbiterium, dobudowaną w 1902 r. wieżą oraz oryginalnym drewnianym sufitem kasetonowym. W 1710 roku pożar zniszczył kościół, a w 1722 roku na polecenie biskupa Macieja Ancuty został odbudowany. Bardzo prawdopodobna jest informacja o sarkofagu z sercem króla Zygmunta Augusta, który znajduje się w podziemiach kościoła. Natomiast w środku kościoła można zobaczyć tablicę pamiątkową, na której umieszczone są słowa króla "Nie jestem królem waszych sumień". Słowa te świadczą o wielkiej mądrości króla, któremu przyszło władać państwem bardzo różnorodnym pod względem narodowym i religijnym. Ostatni król z dynastii Jagiellonów, Zygmunt August miał wielkie plany związane z Knyszynem.
