W 1632 r. mieszczanin bielski Marcin Bartoszewicz sprowadził do Bielska zakonników zgromadzenia karmelitów trzewiczkowych. W 1784 r. w czasie pożaru miasta ogień strawił drewniany kościół oraz częściowo zabudowania klasztorne. Nową świątynię odbudowano w stylu neoklasycystycznym z portykiem na fasadzie. W 1802 r. władze pruskie przeniosły bielskich zakonników do zgromadzenia karmelitów w Wąsoszu. Opustoszałe budynki klasztorne przejęto na cele wojskowe i więziennictwa. W latach 1803-1838 kościół użytkowali unici z parafii Zmartwychwstania Pańskiego. Od lat 40. ych XIX w. pełnił on funkcję prawosławnej cerkwi garnizonowej, w 1866 r. został zaś poświęcony jako sobór św. Trójcy. W tym okresie świątyni nadano wygląd neobizantyjski: wzniesiono dwie wieże z kopułami na frontonie oraz kopułę nad częścią ołtarzową. W 1921 r. kościół został zwrócony prawowitym właścicielom.
